Analysis of Expressed Sequence Tags from the Fungal Banana Pathogen Mycosphaerella fijiensis

چکیده
Mycosphaerella fijiensis is a fungal pathogen causing black leaf streak disease in banana plants. In order to develop our understanding of the molecular mechanisms of host infection by M. fijiensis, we initiated a first step towards large-scale gene discovery through the generation and analysis of expressed sequence tags (ESTs). Three cDNA libraries representing different culture conditions were constructed. A subset of clones was sequenced at the 5’ end and a total of 3,771 ESTs were generated. These ESTs were clustered into a set of 1,945 unique sequences. BLASTX analysis indicated that 410 of the unisequences showed homology to genes with known function, including those related to pathogenicity and virulence. Meanwhile, 66% and 89% of the ESTs matched to the predicted transcripts and genome sequences of M. fijiensis, respectively. The diversity of the EST collection developed in this study will facilitate genome annotation as well as identification and characterization of genes involved in host infection and fungal development through a comparative and functional genomics approach.

Identification of Differentially-Expressed Genes in Response to Mycosphaerella fijiensis in the Resistant Musa Accession ‘Calcutta-4’ Using Suppression Subtractive Hybridization
Bananas and plantains are considered an important crop around the world. Banana production is affected by several constraints, of which Black Sigatoka Disease, caused by the fungus Mycosphaerella fijiensis, is considered one of the most important diseases in banana plantations. The banana accession 'Calcutta-4' has a natural resistance to Black Sigatoka; however, the fruit is not valuable for c...
متن کاملVariable number of tandem repeat markers in the genome sequence of Mycosphaerella fijiensis, the causal agent of black leaf streak disease of banana (Musa spp).
We searched the genome of Mycosphaerella fijiensis for molecular markers that would allow population genetics analysis of this plant pathogen. M. fijiensis, the causal agent of banana leaf streak disease, also known as black Sigatoka, is the most devastating pathogen attacking bananas (Musa spp). Recently, the entire genome sequence of M. fijiensis became available. We screened this database fo...

Fungi are the causal agents of many of the world's most serious plant diseases causing disastrous consequences for large-scale agricultural production. Pathogenicity genomic basis is complex in fungi as multicellular eukaryotic pathogens. Here, we report the genome sequence of C. sojina, and comparative genome analysis with plant pathogen members of the genus Mycosphaerella (Zymoseptoria. triti...
متن کاملSinglet Molecular Oxygen Generation by Light-Activated DHN-Melanin of the Fungal Pathogen Mycosphaerella fijiensis in Black Sigatoka Disease of Bananas
In pathogenic fungi, melanin contributes to virulence, allowing tissue invasion and inactivation of the plant defence system, but has never been implicated as a factor for host cell death, or as a light-activated phytotoxin. Our research shows that melanin synthesized by the fungal banana pathogen Mycosphaerella fijiensis acts as a virulence factor through the photogeneration of singlet molecul...

Black Sigatoka or black leaf streak disease, caused by the Dothideomycete fungus Pseudocercospora fijiensis (previously: Mycosphaerella fijiensis), is the most significant foliar disease of banana worldwide. Due to the lack of effective host resistance, management of this disease requires frequent fungicide applications, which greatly increase the economic and environmental costs to produce ban...
